Mayo Clinic finds experimental drug inhibits growth in all stages of common kidney cancer

JACKSONVILLE, Fla. Researchers at Mayo Clinic's campus in Florida have discovered a protein that is overly active in every human sample of kidney cancer they examined. They also found that an experimental drug designed to block the protein's activity significantly reduced tumor growth in animals when used alone. Combining it with another drug already used to treat the cancer improved the effectiveness of both.
The findings, reported in the April 30 online issue of Clinical Cancer Research, offer a much-needed potential new direction for the treatment of clear cell renal cell carcinoma, which accounts for almost 85 percent of kidney cancer cases in the United States. More than 57,000 diagnoses of kidney cancer occur yearly in the U.S. with greater than 13,000 deaths.
"There is a clear need for new therapies for this common cancer. With very few exceptions, patients inevitably become resistant to all available treatments," says the study's senior investigator, molecular biologist John Copland, Ph.D.
Their findings might be relevant to the treatment of other cancers, says Christina von Roemeling, the study's lead author. The protein they identified is produced by the stearoyl CoA desaturase 1 (SCD1) gene, which has also been found to be over active in a number of other cancers, including lung, stomach, breast, prostate, ovary and colon cancers.
"This is a gene that is highly active in a lot of other cancers and it may be that the agent we tested could provide new clinical avenues in those cancers as well," von Roemeling says.
The experimental drug, A939572, is a targeted inhibitor of SCD1 protein. "We found it to be incredibly specific to cancer cells in laboratory mice treated with the agent," Dr. Copland says. "But these are early days in the testing of this agent for cancer."
Not only is SCD1 active in some cancers, it also is being investigated for its role in promoting obesity and diabetes, the researchers say. Scientists are also testing A939572 as an antidote to those conditions.
The Mayo Clinic scientists performed a genome screen of tissue samples from 150 kidney cancer patient tissue samples, which represented all stages of cancer progression, to identify genes that are significantly overexpressed, compared to noncancerous tissue samples. SCD1 was one of their top finds.
They then disabled SCD1 in laboratory kidney cancer cells and found that the tumor cells stopped growing and a large percentage died.
Next, researchers tested A939572 and the federally approved kidney cancer drug temsirolimus. They found that using either agent alone cut tumor growth by up to 25 percent in mice studies, but using both drugs together, and at lower doses, reduced it 60 to 70 percent.
"The synergy between the drugs was very striking, suggestive of significant clinical benefit in patients," Dr. Copland says.
Von Roemeling says that SCD1 protein expression offers a novel molecular prognostic biomarker in kidney cancer that could guide therapy.

Lawmakers: Syria chemical weapons could menace U.S.
WASHINGTON (AP) ? Syria's stockpile of chemical weapons could be a greater threat after that nation's president leaves power and could end up targeting Americans at home, lawmakers warned Sunday as they considered a U.S. response that stops short of sending military forces there.
U.S. officials last week declared that the Syrian government probably had used chemical weapons twice in March, newly provocative acts in the 2-year civil war that has killed more than 70,000 people and displaced hundreds of thousands more. The U.S. assessment followed similar conclusions from Britain, France, Israel and Qatar ? key allies eager for a more aggressive response to the Syrian conflict.
President Barack Obama has said Syria's likely action ? or the transfer of President Bashar Assad's stockpiles to terrorists ? would cross a "red line" that would compel the United States to act.
Lawmakers sought to remind viewers on Sunday news programs of Obama's declaration while discouraging a U.S. foothold on the ground there.
"The president has laid down the line, and it can't be a dotted line. It can't be anything other than a red line," said House Intelligence Committee Chairman Rep. Mike Rogers, R-Mich. "And more than just Syria, Iran is paying attention to this. North Korea is paying attention to this."
Added Sen. Saxby Chambliss, R-Ga.: "For America to sit on the sidelines and do nothing is a huge mistake."
Obama has insisted that any use of chemical weapons would change his thinking about the United States' role in Syria but said he didn't have enough information to order aggressive action.
"For the Syrian government to utilize chemical weapons on its people crosses a line that will change my calculus and how the United States approaches these issues," Obama said Friday.
But Rep. Jan Schakowsky, an Illinois Democrat, said Sunday the United States needs to consider those weapons. She said that when Assad leaves power, his opponents could have access to those weapons or they could fall into the hands of U.S. enemies.
"The day after Assad is the day that these chemical weapons could be at risk ... (and) we could be in bigger, even bigger trouble," she said.
Both sides of the civil war already accuse each other of using the chemical weapons.
The deadliest such alleged attack was in the Khan al-Assal village in the Aleppo province in March. The Syrian government called for the United Nations to investigate alleged chemical weapons use by rebels in the attack that killed 31 people.
Syria, however, has not allowed a team of experts into the country because it wants the investigation limited to the single Khan al-Assal incident, while U.N. Secretary-General Ban Ki-moon has urged "immediate and unfettered access" for an expanded investigation.
One of Obama's chief antagonists on Syria, Sen. John McCain, R- Ariz., said the United States should go to Syria as part of an international force to safeguard the chemical weapons. But McCain added that he is not advocating sending ground troops to the nation.
"The worst thing the United States could do right now is put boots on the ground on Syria. That would turn the people against us," McCain said.
His friend, Sen. Lindsey Graham, R-S.C., also said the United States could safeguard the weapons without a ground force. But he cautioned the weapons must be protected for fear that Americans could be targeted. Raising the specter of the lethal bomb at the finish line of the Boston Marathon, Graham said the next attack on U.S. soil could employ weapons that were once part of Assad's arsenal.
"Chemical weapons ? enough to kill millions of people ? are going to be compromised and fall into the wrong hands, and the next bomb that goes off in America may not have nails and glass in it," he said.
Rogers and Schakowsky spoke to ABC's "This Week." Chambliss and Graham were interviewed on CBS's "Face the Nation." McCain appeared on NBC's "Meet the Press."

Jeter has 'no doubt' that he'll return to Yankees
NEW YORK (AP) ? Don't count out the captain.
Derek Jeter is certain he will return to the New York Yankees this season at the same level that made him one of baseball's most revered players.
In a jocular mood and wearing pinstriped pants, a practice jersey and hat, Jeter expressed frustration in learning last week that there was a new break in the ankle, an injury that has sidelined him since Game 1 of the AL championship series. He likely will be out until after the All-Star break.
Still, he never wavered in his confidence that he will run ? without a limp ? to the shortstop position that has been his since 1996.
"When you have doubt, that's when you're in trouble. I have been told this bone will heal, and when it heals I'll be ready to go," Jeter said Thursday before the Yankees' 5-3 win over Toronto. "It's frustrating I can't magically make it heal sooner than it's taken."
Jeter was in the dugout for a game for the first time this season, getting to know several teammates that weren't with the Yankees when he played in his last spring training game in mid-March.
Kevin Youkilis, among the new Yankees, was out of the lineup for the fifth straight game because of a tight lower back.
"He tried to take some work in the cage," manager Joe Girardi said. "He's just not ready. I thought it would be today. So hopefully it's tomorrow. We'll just go day by day."
Girardi is confident that when Jeter returns, he'll be the same player who has 3,304 hits, including an AL-leading 216 last season.
"He's had a setback here," Girardi said. "We have to deal with it but, hopefully, we get him back and he's the same player he was at the end of last year."
The 13-time All-Star is disappointed he failed in fulfilling his prediction that he would return by opening day. Now the reality is he will not be able to help his team until around mid-July, weeks after he's turned 39 years old.
No. 2 has a date in mind for his return, but he not saying when it is.
"The last timeline I set, I didn't make," Jeter said. "I don't want to disappoint myself or anyone else."
Jeter was always stubborn about injuries, refusing tests and claiming he was well enough to play. He has willed himself onto the field throughout a career in which his 2,531 games at shortstop rank only behind Omar Vizquel (2,709) and Luis Aparicio (2,581).
"I don't talk about injuries," he said. "It's just I think talking about injuries is just making an excuse for yourself. You either play or you don't."
In 2004, he famously dived into the stands to make a catch against the Boston Red Sox and walked off the field bloody and bruised. Yet, he took his position at Shea Stadium the next day.
When he dislocated his shoulder in 2003, he returned almost exactly to the day predicted and played through pain much of the season ? the only one of his 17 previous full seasons that he played less than 130 games.
At 38, he had no such luck. Jeter played for much of last September with a bad bone bruise. It finally gave out against the Detroit Tigers when he lunged for a groundball Oct. 13. A week later he had surgery, and a Christmas party at Yankee Stadium for his Turn 2 Foundation, he vowed to be on the field for the April 1 opener.
But Jeter was slowed by stiffness and soreness during spring training and only played five big league games, three at shortstop. When the pain persisted into April, he went for a new CT scan in Charlotte, N.C., and that test revealed the break.
"When I got it, it wasn't good news," Jeter said. "I thought I would go up there, when I went to see the doc, I thought he would say it was something different. Tape it up. Let's go. But it wasn't the case. It didn't feel too good for quite some time. I'm laughing and smiling and happy that I'm up here. But I'm still upset that I can't play."
For now he will be limited to playing cheerleader and working out until he is given the OK to start his on-field rehabilitation again. Jeter walked without a limp into and out of the news conference, and he wasn't wearing the protective boot he says he has to wear, even though he doesn't think it's necessary.
He'll spend at least the 10-game homestand in New York.
"It's tough to not be around the team," Jeter said.

ECB: lending to companies remains weak
FRANKFURT, Germany (AP) ? The European Central Bank says bank lending to companies remains weak ? another sign that the economy in the 17 countries that use the euro is still stuck in recession.
Loans to companies shrank 2.4 percent in March from the same month a year ago, only slightly better than the drop of 2.5 percent the month before.
The ECB says its current low interest rates are not getting through to companies in the form of credit that would enable them to expand their businesses. That's because of troubles in the banking system in some parts of the eurozone.
Many economists think the ECB will cut its refinancing rate from a record low of 0.75 percent at next week's meeting to provide more stimulus.

Have no fear, there is plenty of great talent remaining on day two. Expect a large run on quarterbacks, wide receivers, tight ends, and defensive backs during the second- and third-rounds. There?s a variety of reasons the prospects listed below missed out on the first-round. Some possibly due to medicals (Brown, Carradine), others a poor pre-draft process (Allen, Lacy), and finally, a few specialize in a certain role (Washington, Rambo).
I have not included quarterbacks, but you can find my full list here. I?ve narrowed the evaluation of each of these prospects down to what truly matters: where they can help the team that drafts them. Next to each name is where they ranked on my pre-draft top 151.
1. LB Arthur Brown, Kansas State (8)
Where he wins: Instinctive, quick, and physical for his size. Believe he could play inside in any scheme but could thrive on the weakside. Doesn?t shy away from contact but also has the speed to beat ball carriers to the edge or stick with his assignment in coverage.

Where he wins: Explosive off the snap and follows it up with strong hand use to keep separation or disengage. Plays with leverage to drive opposition back if need be or keeps his position to set the edge as a run defender.

NEW YORK (AP) ? Scientists have identified a hormone that can sharply boost the number of cells that make insulin in mice, a discovery that may someday lead to a treatment for the most common type of diabetes.
People have their own version of this hormone, and the new work suggests that giving diabetics more might one day help them avoid insulin shots.
That would give them better control of their blood sugar levels, said Harvard University researcher Douglas Melton, senior author of a report published Thursday by the journal Cell.
Experts unconnected with the work cautioned that other substances have shown similar effects on mouse cells but failed to work on human ones. Melton said this hormone stands out because its effect is unusually potent and confined to just the cells that make insulin.
An estimated 371 million people worldwide have diabetes, in which insulin fails to control blood sugar levels. High blood sugar can lead to heart disease, stroke and damage to kidneys, eyes and the nervous system. At least 90 percent of diabetes is "Type 2," and some of those patients have to inject insulin. Melton said the newly identified hormone might someday enable them to stop insulin injections and help other diabetic patients avoid them.
As for its possible use to treat Type 1 diabetes, Melton called that a "long shot" because of differences in the biology of that disease.
Insulin is produced by beta cells in the pancreas.
Melton and co-authors identified a hormone they dubbed betatrophin (BAY-tuh-TROH-fin) in mice. When they made the liver in mice secrete more of it by inserting extra copies of the gene, the size of the beta cell population tripled in comparison to untreated mice. Tests indicated the new cells worked normally.
Melton said it's not known how the hormone works. Now the researchers want to create an injectable form that they can test on diabetic mice, he said. If all goes well, tests in people could follow fairly quickly.
Dr. Peter Butler, a diabetes researcher at the University of California, Los Angeles, who had no role in the new work, cautioned in an email that no evidence has been presented yet to show that the hormone will make human beta cells proliferate.
But Philip diIorio, of the University of Massachusetts Medical School in Worcester, said he found the work to be "quite promising" because it offers new leads for research, and that it might someday help in building supplies of human beta cells in a lab for transplant into patients.

U.S. government, Fisker grilled over loans to green carmaker

(Reuters) - Taxpayer-backed funds kept flowing to electric carmaker Fisker Automotive months after the company failed to meet key production benchmarks, lawmakers said at a congressional hearing on Wednesday.
Republican lawmakers on the House Oversight Committee cited Department of Energy documents as showing Fisker got $32 million in payments, even after it failed to launch its Karma vehicle in February of 2011.
They spent hours quizzing current and former Fisker executives and an Energy Department official over what the lawmakers termed the government's "bad bet" on the fledgling luxury car-maker, and questioned whether the company received special treatment that put taxpayer dollars at risk.
The committee hearing focused on the DOE's decision in 2009 to grant the company a $529 million loan only to see it veer toward bankruptcy - a chain of events with echoes of Solyndra, the government-backed solar manufacturer that went out of business in 2011.
"The government is a very poor venture capitalist," said Patrick McHenry, a Republican from North Carolina.
Fisker's troubles are the latest in a string of green automotive technology flops, including last year's bankruptcy of its lithium-ion battery supplier, A123 Systems. Forecasts in 2009 for sales of hybrid and electric vehicles far outstripped subsequent demand.
Henrik Fisker, founder and former CEO, said the company, which has not built a vehicle since July, can still bounce back and repay nearly $200 million in government loans if is able to find the right "financial and strategic resources."
Testifying about his eponymous company, the Danish-born Fisker, who was forced to resign as chairman in March, blamed problems with its parts suppliers, delays in regulatory approval and recalls of its flagship Karma plug-in hybrid sports car for the company's struggles.
The automaker verges on collapse. Among the questions still outstanding is whether Fisker's prospects were ever strong enough to warrant the DOE's backing, which helped trigger a flood of private financing for Fisker.
"After resolving initial launch challenges, the cars perform well and customers love them," Fisker asserted.
Fisker's failure to make a payment on the DOE loan on Monday was the latest of its troubles. In recent weeks, Fisker has fired 75 percent of its workforce and hired bankruptcy advisers.
"The Obama Administration owes the American taxpayer an explanation as to why this bad loan was made in the first place, and what they are going to do to minimize the loss that taxpayers face," said Ohio Republican Jim Jordan, chairman of the subcommittee holding Wednesday's hearing.
DOE BACKING BOOSTED FISKER
The DOE's early backing helped put Fisker, and its $100,000-plus Karma plug-in hybrid sports car, on the map. With government backing in hand, Fisker has gone on to raise $1.2 billion in private funds to date, according to SEC filings.
The 2009 loan signaled that the DOE had done a rigorous review of the project, said Salo Zelermyer, a senior counsel at the DOE under the Bush administration, who also helped create the auto loan program. The loan program was funded in late 2008.
"It's fair to say the projects the DOE chose to proceed with were clearly given an added credibility with folks on the outside," said Zelermyer, now a senior counsel at Bracewell and Giuliani in Washington.
Fisker tapped $192 million of its $529 million before the DOE quietly decided to freeze Fisker's credit line in June 2011.
Neither the DOE nor Fisker publicly disclosed that decision until early 2012. Lawyers and a DOE official said the department was not obligated to divulge the decision.
In the confidential "information statement" sent to shareholders in December 2011 and obtained by Reuters, Fisker said it "will not meet certain financial covenants and project milestones" required in the DOE agreement, including earnings, net worth and certain financial ratio targets.
Lawmakers raised questions about the relationship between the DOE and Fisker, which has been strained in recent months, people familiar with the matter have said.
Republicans said Fisker received loans despite its junk bond rating and unproven track record.
The terms of Fisker's pact with the DOE were enough to put off potential suitors, including Chinese automaker Geely Automobile Holdings Ltd. The conditions included an obligation to restore capacity and jobs at the company's Delaware plant according to a schedule imposed by the U.S. government.
Energy Department officials defended the auto loan program, saying it helped to bring the industry from the brink of collapse during a severe economic downturn. Mainstream carmakers like GM and Chrysler were key beneficiaries, and their revival has been widely acclaimed.
Nicholas Whitcombe, former head of the auto loan program, said the DOE remains open to providing additional aid to the auto industry.
Since the high profile failure of Solyndra, a topic brought up regularly by Republicans during President Barack Obama's 2012 re-election campaign, the Obama administration has become far more risk averse when doling out loan payments.
Solyndra, the first loan recipient and first major failure for the department's portfolio, received more than $527 million of its $535 million loan before filing for bankruptcy.

American-Norwegian freeskier Grete Eliassen has received the most number of votes in ESPN?s Best Female in Action Sports poll.
Voting remained opened between March 19 and April11, during which action sports enthusiasts voted feverishly for their favourite action sports athletes with the intended motivation of making them the winner.
Among the many categories of polls and brackets set up by ESPN was the poll for the most notable female athletes in ESPN. The options included the likes of five-time Association of Surfing Professionals (ASP) Women?s World Champion Stephanie Gilmore, her fellow surfer Coco Ho, skaters Mimi Knoop and Leticia Bufoni, snowboarder Jess Kimura.
The poll attracted 54,166 votes, 64 percent of which went to Eliassen, thus making her a winning by a comprehensive margin.
The freeskier was surprised at managing to finish well above the top athlete sports female athletes and made no effort to hide her surprise and excitement.
She said that once she was informed about being in a bracket by her brother and that she was losing, she took to social media and got her family, friends and fans involved.
?I was totally surprised. My brother told me I was in a bracket and I was losing, so I put some stuff on my social media sites and called my family and started some momentum. It just carried on through and I won. To be on the same level as Travis Rice is pretty cool. I couldn't have done it without the fans. My Facebook followers don't feel like random strangers anymore. It feels like a team,? she admitted during an exclusive interview with ESPN.
Eliassen has gained prominence in action sports, especially after putting on a sensational performance to clinch bronze medal in Freeskiing Slopestyle at Winter X Games 2011 at Aspen, Colorado, losing the top two podium places to Kaya Turski and Keri Herman.
She tore her ACL last year and was unable to compete in the most recent season, but the Norwegian-American freeskier is all fired up to hit the slopes and halfpipe again.
With the Winter Olympics not too far away, she would surely be looking to build some momentum by participating in as many freestyle skiing competition as possible and trying to make a solid impact, if not win, in as many of them as possible.

Fake tweet shows country 'sensitive to any news that sounds like terrorism'

A stock market and a nation already on edge was temporarily knocked off its axis on Tuesday by a single fake tweet.?
Following a hack attack, the Associated Press' verified Twitter account posted "an erroneous tweet" claiming that two explosions occurred in the White House and that President Barack Obama is injured. Moments later, the @AP Twitter account ? with nearly 2 million followers ? was suspended.
"That's a bogus tweet," an AP spokesperson initially told NBC News, a statement that was?repeated by the company's corporate communications account. Though the false tweet disappeared, the false message continued to exist on the service in over 4,000 retweets.
The chart of the Dow Jones industrial average just after 1 p.m. may as well have been a chart of America's heartbeat -- stopped for a moment, again, by seemingly horrific information. The Dow lost more than 140 points almost instantly, before recovering five minutes later.
It's incredible what a single 12-word lie can do.
The markets plummet, and then snap back after a fake AP terror tweet, with the "Power Lunch" crew.
"We're in an environment where we're sensitive to any news that sounds like terrorism," said Art Hogan of Lazard Capital Markets.? "That makes it that much more believable. That's the tricky part. When something like AP gets hacked, it becomes reality for a period of time, until it's not."
The market's reaction hints at the our collective fragility right now.? In the past, carefully crafted fake press releases or other Internet disinformation has been able to influence individual stocks both up or down.
But a single Tweet sinking the market?? It's just the latest sign that lies now spread on the Internet as fast as computer viruses, and can have just as much impact. Like the false rumors that spread like wildfire during the Boston bombing aftermath, or Hurricane Sandy before that, Twitter's surge to mainstream popularity ? it now boasts 140 million U.S. accounts???has made it an incredible source of on-the-spot information, but also the world's most powerful rumor-mongering tool.
"You wonder who did it and whether it was done on purpose. It certainly was an instant implosion," said Art Cashin, director of floor operations for UBS Financial Services, who watched the minutes of bedlam on the floor of the NYSE. Cashin said the reaction was especially dramatic because it said the president was injured.
If you define the term "hacking" loosely, you might consider that whoever wrote the fake tweet hacked not only AP's account, but the entire Wall Street trading system. The trades which sank the market Tuesday were almost certainly initiated by automated trading programs designed to profit by fast-twitch reacting to good or bad news.
The combination of a jittery public, automated trading, and a worldwide rumor tool was toxic for the markets.
"That goes to show you how algorithms read headlines and create these automatic orders ? you don't even have time to react as a human being," said Kenny Polcari of O'Neil Securities. "I'd imagine the (Security and Exchange Commission) is going to look into how this happened. It's not about banning computers, but it's about protection and securing our markets."
It's also about figuring out how to handle a world where the firewall between seemingly disconnected systems like Twitter and brokerage servers is really only 91 characters long, particularly a world where skepticism?s classic grains of salt seem to be in short supply.
